I. Background | |
激活素 A(Activin A)是轉(zhuǎn)化生長因子-β(TGF-β)超家族的重要成員,是一種多能因 子,參與過敏、自身免疫性疾病、癌癥等免疫失調(diào)疾病的發(fā)生。 激活素 A 通過兩種 I 型受體和兩種 II 型受體發(fā)出信號(hào),當(dāng)它們與配體結(jié)合時(shí),組裝成 最終的受體復(fù)合物。I 型受體包括 1A 型激活素受體(或激活素受體樣激酶 2,ALK2)、 1 B 型激活素受體(或 ALK4)和 1C 型激活素受體(或 ALK7)。Activin-A 有利于 ALK4 結(jié) 合,而對(duì) ALK2 和 ALK7 的親和力較低。II 型受體是 Activin 受體 IIA 型(ActRIIA)和 Act ivin 受體 IIB 型(ActRIIB),其特征是組成性活性絲氨酸/蘇氨酸激酶活性。一旦 Activin A 與其 II 型受體結(jié)合,兩個(gè) I 型受體就會(huì)被募集,并被 II 型受體磷酸化,這一事件導(dǎo)致其激 酶活性被激活。大多數(shù) TGF-β超家族成員與 ActRIIB 或 ActRIIA 結(jié)合,而只有少數(shù)配體與 ALK4 高親和力結(jié)合。除了典型受體絲氨酸/蘇氨酸激酶- smad 軸外,激活素 A 還被證明參 與 P38、ERK、PI3K、WNT 等途徑。 |
